Abstract

Subject. Enzymes and inhibitors are unstable to the action of external factors (temperature, pH), therefore the question of their stabilization by immobilization on biomatrices arises. The study of immobilization using IR spectroscopy allows us to speak more accurately about the process of interaction of enzyme inhibitor and biomatrix. Purpose. Study of the kinetic characteristics of a biocomplex based on pancreatic amylase inhibitor and collagen hydrolyzate. Methods. Determination of amylase enzyme activity and the influence of the immobilized form of the inhibitor on it. Determination of bonds when obtaining a biocomplex using IR spectroscopy and the nature of the inhibition kinetics of linearization of the Michaelis-Menten equation according to Lineiver-Burke and Haynes. Results. Using IR spectroscopy, it was shown that the crosslinking of the amylase inhibitor with the biomatrix occurs through the formation of intramolecular covalent bonds, electrostatic interaction between the charged groups of collagen hydrolysate and amylase inhibitor. The differential comparison curve of the VVOP is characterized by a decrease in the absorption intensity of the immobilized inhibitor in the region of 3400 cm-1, which corresponds to the valence vibrations of the free -OH group, which indicates the strengthening of hydrogen bonds of the immobilized sample. Kinetic parameters of amylase inhibition were performed using the linearization methods of the Michaelis-Menten equation according to Lineiver-Burke and Haynes. The study of the kinetics of amylase inhibition by the immobilized form of the inhibitor showed a decrease in enzymatic activity, characterized by the absence of changes in Km with a decrease in the Vmax value, which allows us to attribute the inhibition to the linear non-competitive type. Scope of results. Biotechnology of obtaining biocomplexes capable of regulating nutritional processes.
